Artesunate (ART), a semi-synthetic derivative of the sesquiterpene artemisinin extracted from the Chinese herb
Artemisia annua, exerts a broad spectrum of clinical activity against human cancers. It has been shown that ART induces
cancer cells death through apoptosis pathway. This study investigated whether ART treatment induced
reactive oxygen species (ROS)-dependent cell death in the apoptosis fashion in human lung adenocarconoma A549 cell
line and the proapoptotic protein apoptosis inducing factor (AIF) is involved in ART-induced apoptosis. Cells treated
with ART exhibited typical apoptotic morphology as chromatin condensation, margination and shrunken nucleus. ART
treatment also induced a loss of mitochondrial membrane potential and AIF release from mitochondria. Silencing AIF
can remarkable attenuated ART-induced apoptosis. Collectively, ART induces apoptosis by
caspase-independent intrinsic pathway in A549 cells.
