At the present time a new direction of theranostics associated with using of nanostructured multifunctional carriers is developed rapidly. Appreciative examples of such carriers are core-shell nanoparticles, liposomes, polymer micelles, capsules produced by Layer by Layer assembly (LbL) method. Every type of nanostructured carriers has its own advantages and disadvantages. Undoubted advantages of LbL capsules are high loading capacity, wide range of possible internal payloads and wide possibilities of controlling physical and chemical properties of nanostructured carriers by variation of shell structure, thickness and its chemical composition [М.Delcea, et al, Adv. drug delivery reviews, 2011, 63(9), 730, A.S.Timin, et al, Adv. Mater. Interfaces,2016, 4, 1600338]. Application of nanostructured carriers is limited by absence of methods for its in vivo visualization with sufficient spatial resolution and significant tissue penetration depth [A.S.Timin, et al, Adv. Mater. Interfaces, 2016, 4,1600338]. One of the method that allows to visualize and to detect the nanostructured carriers efficiently is a photoacoustic (PA) method. PA approach allows to use the photoacoustic cytometry in vivo [E.I. Galanzha et al, Cancer Res. 2009, 69, 7926]. It is also very important for detection and killing of free circulating cancer cells required for early cancer diagnostics and decreasing the metastases probability [E.I. Galanzha, et al, J. Biophotonics, 2009, 2, 725]. Unique combination of photoacoustic cytometry and nanostructured carriers prepared by LbL assembly approach gives clinicians and researchers significant advantages in the developing of new therapy method based on delivery of nanostructured carriers loaded by bioactive substances into lesion focus and also their visualization and remote controlled release of encapsulated bioactive substances using PA approach. Visualization of two types of nanostructured carriers produced by LbL assembly method was demonstrated recently in diluted and undiluted blood [E.I. Galanzha, et al, J. Biophotonics, 2009, 2, 725]. It was established that hollow microcapsules exhibited greater photoacoustic signal comparing to core-shell type of microparticles with the same composition of polymeric shell [E.I. Galanzha, et al, J. Biophotonics, 2009, 2, 725]. Nevertheless, there are no known examples of in vivo photoacoustic detection of LbL nanostructured carriers, therefore the main goal of present study is formation of biocompatible nanostructured carriers (BNCs) that will exhibit giant photoacoustic signal allowing in vivo detection of such structures. It was demonstrated for the first time, the super (giant) PA contrast of BNCs both in vitro and in vivo. It was verified the unique PA characteristics of these advanced contrast agents using PA flow cytometry diagnostic platform. The obtained data suggest the high PA contrast of BNCs that can be associated with synergistic plasmonic, thermal and acoustic effects, especially in nonlinear mode with nanobubble formation in overheated absorbing layers, in particular gold nanoclusters between two light transparent shells. Background and Objective: The application of nanotechnology for laser thermal-based killing of abnormal cells (e.g. cancer cells) targeted with absorbing nanoparticles (e.g. gold solid nanospheres, nanoshells, or rod) is becoming an extensive area of research. We develop an approach to enhance the efficiency of selective nanophotothermolysis of cancer cells through laser-induced synergistic effects around gold nanoparticles aggregated in nanoclusters on cell membrane. Study Design/Materials and Methods: A concept of selective target damages by laser-induced synergistic interaction of optical, thermal, and acoustic fields around clustered nanoparticles is presented with focus on overlapping bubbles from nanoparticles aggregated on cell's membrane. The experimental verification of this concept in vitro was performed by the use a tunable laser pulses (420-570 nm, 8-12 ns, 0.1-300 μJ, laser flux of 0.1-10 J/cm²) for irradiation of MDA-MB-231 breast cancer cells targeted with primary antibodies to which selecttively 40-nm gold nanoparticles were attached by the means of secondary antibodies. The photothermal, electron and atomic force microscopes in combination with viability test (annexin -V-Propidium iodide) were employed to study the nanoparticle's spatial organization, the dynamics of microbubble formations around the particle's clusters, and cells damage. Results: An aggregation of nanoparticles on cell membrane was observed with simultaneous increase bubble formation phenomena, and red-shifted absorption due to plasmon-plasmon resonances into nanoclusters. It led to a significant enhancement, at least two orders of magnitude, of the efficiency of selectively killing cancer cells with nanosecond laser pulses. Conclusion: Described approach allows using relatively small nanoparticles which would be easier delivery to target site with further creation of nanoclusters with larger sizes which provide more profound thermal and related phenomena leading to more efficient laser killing of cancer cells. This nanocluster model might be promising also for treatment or modification different targets (e.g. bacteria, virus, vascular lesions, fat, etc.) as well as teh use different type energy deposition (focused ultrasound, microwave, magnetic field, etc.).

This short review presents findings from a recent evaluation of the diagnostic capabilities of a new experimental design of the advanced photothermal (PT) imaging system; specifically, its performance in studying the impact of nicotine, a combination of antitumor drugs, and radiation on the absorbing structures of various cells. We used this imaging system to test our hypothesis that low doses of chemicals or drugs lead to changes in cell metabolism, that these changes are accompanied by the shrinking of cellular absorbing zones (e.g. organelles), and that these reactions cause increased local absorption. Conversely, high (toxic) doses may lead to swelling of organelles or release of chromophores into the intracellular space, causing decreased local absorption. In this study, we compared PT images and PT responses of the pancreatic exocrine tumor cell line AR42J resulting from exposure to various concentrations of nicotine versus those of control cells. We found that responses were almost proportional to the drug concentration in concentrations ranging from 1 nM-100 μM, reached saturation at a maximum of approximately 100 μM-1 mM, and then fell rapidly at concentrations ranging from 1-50 mM. We also examined the influence of antitumor drugs (vinblastine and paclitaxel) on KB3 carcinoma cells, with drug concentrations ranging from 10^-10 nM to 10 nM. In this instance, exposure initially led to slight cell activation, which was then followed by decreased cellular PT response. Drug administration led to corresponding changes in the amplitude and spatial intracellular localization of PT responses, including bubble formation, as an indicator of local absorption level. Additionally, it was shown that, depending on cell type, x-ray radiation may produce effects similar to those resulting from exposure to drugs. Independent verification with a combined PT-fluorescence assay and conventional staining kits (trypan blue, Annexin V-propidium iodide [PI]) revealed that this new PT assay has the potential to detect different stages of environmental impact, including changes in cell metabolism and apoptotic- and toxic-related phenomena, at a concentration threshold sensitivity at least three orders of magnitude better than existing assays. This assay may also help optimize combined cancer therapies.

Photothermal method was applied to improve sensing and imaging capabilities of a light microscope in cell studies. We describe the methods, technical details and testing results of cytometric application of Laser Photothermal Phase Microscope (LPPM). The merits of the proposed approach include living single cell monitoring capability, quantitative measurement of cell functional features through the use of cell natural chromophores as the sensors. Such intracellular sensors are activated by the laser pulse and transform an absorbed energy into the heat. The latter causes thermal and mechanical loads to a cell and its components. The second stage of the process includes the reaction of the cell as integral system or of its components to such loads. This reaction is caused by the changes of cell functional and structural state and includes alterations of cell optical properties. Both processes are monitored for a single cell non-invasively with probe laser beam. Pulsed phase contrast dual beam illumination scheme with acquisition of several laser images at different stages of cell-laser interaction was introduced. An acquired cell image is considered as spatially and temporally resolved cell response to non-specific load that is induced in a cell with a pump laser. This method eliminates any cell staining and allows to monitor cell viability and cell reaction to the environmental factors. Also LPPM offers further improvement of spatial and temporal resolution of optical microscope: with pulsed probe laser monitoring we can detect components with the size down to 50 nm and temporal resolution of 10 ns. In our set up the cell is pumped by pulsed laser at 532 nm, 10 ns , 0.01-0.4 mJ. The source of probe beam is a pulsed dye laser (630 nm, 10 nJ, 10 ns) which forms cell phase image. The results obtained with living cells such as drug impact control, single cell dosimetry, immune action of light on a cell demonstrate basic features of LPPM as the tool for the study of the spectral and spatial low-absorption distribution in living cells in addition to techniques of active thermal probing of sub-micron, low-absorbing samples.

According to the world statistics, from 30 to 60 percent of elderly male population suffer from chronic prostatitis in different countries. This disease has a number of consequences such as urino-genital inflammation, dysuria, perineal pain, reduction in the physiological activity of smooth muscles, blockage of the anus passages with micro-organism vital activity products, appearance of stagnant zones and low blood circulation complicated by disorders of the sexual function. Most of these features make it difficult to use standard drug therapies with antibiotics or immunocorrectors. For that reason, the objective of this study is to develop and to investigate a novel combined electrolaser therapy which improves drug delivery in the prostate gland and simultaneously provides an independent physiotherapeutic effect. The main feature of this therapy is the utilization of two diode lasers emitting in the red (0.67 micrometer, 10 mW) and in the infrared (0.85 micrometer, 1 W) spectrum ranges in combination with transurethral electrostimulation. An electrolaser catheter containing both hollow cylindrical electrodes and an axial optical fiber to deliver laser radiation was brought along the urethra to the seminal vesicles. The red laser in combination with a photosensitizer ('Photosens,' Russia) was used to realize the antibacterial treatment of the urethra. The infrared laser was employed to heat the prostate gland and to stimulate the blood perfusion without thermal damage of tissues. The laser heating of the prostate at a local tissue temperature of 41 degrees Celsius in combination with the electrostimulation provided approximately a 4.5-fold increase in the blood flow. The realization of an additional mode of photovacuum therapy inside the urethra together with the electrostimulation made it possible to 'clean' the anus passages and to improve the DNA diagnosis reliability in respect of the urogenital infectious remainder. The clinical data obtained in 980 patients, who had been undergoing the treatment since 1994 till 1999 in accordance with the electrolaser method in Russia, are analyzed.
